Radical Scavenging Active Compound Analysis in Extract of Humulus japonicus Using an Online LC-DPPH Assay

Abstract
Humulus japonicus is a species that disrupts the ecosystem by interfering with the survival of other plants due to its ability to grow attached to other objects or plants and its prolific reproductive capacity. In order to increase the utilization of H. japonicus, an analysis was conducted on the major antioxidant active compounds present in H. japonicus extract. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured simultaneously with liquid chromatograph (LC) separation, and then UV spectrum measurement and mass spectrometer (MS) analysis were performed for the identified radical scavenging active compound peak. Through literature review and reference standard analysis, it was confirmed that the active compound was luteolin-7-O-glucoside. In quantitative analysis, the content of luteolin-7-O-glucoside in the extract was found to be 0.702 mg/ml, which is about 1.4% of the dried H. japonicus sample. Luteolin-7-O-glucoside is more water-soluble than luteolin, the aglycone of this compound, and has been reported to have various physiological activities, so it should be treated importantly in research using H. japonicus.
